چکیده

A whole variety of chemical and biological materials have immunosuppressive effects in animals. Although anti-inflammatory agents, especially the corticosteroids, are inhibitors of the immune response, most are general cytotoxic agents. The cytotoxic agents were first developed as antitumour agents. It is now well known that they are not particularly selective for cancer cells, but are selectively toxic for cells that are in cycle. Some are even more specific, acting only on cells in a particular phase of the cell cycle. When selective antitumour effects are obtained, this is not through a different mechanism of action, but is a result of other properties of the agent that ensure that a greater reaction takes place in the cancer cell than in sensitive cells of the host. This may be achieved by selective concentration or by selective activation of the agent in the cancer cell. Alternatively the compound may be more readily detoxified by normal cells or these may have a greater ability to repair any damage caused. As a result of their general cytotoxicity these antitumour agents have a number of side effects, most of which are undesirable, but which may sometimes be exploited, such as in their use as immunosuppressive agents. When these agents are used for their immunosuppressive properties their selectivity is once more determined by the properties of the whole molecule, which regulate the amount of agent reacting with the target sites of the immunocyte compared with the amount that reacts with sensitive host cells.

Generation of Epstein-Barr virus-specific cytotoxic T lymphocytes resistant to the immunosuppressive drug tacrolimus (FK506).

Adoptive transfer of autologous Epstein-Barr virus-specific cytotoxic T lymphocytes (EBV-CTLs) to solid organ transplant (SOT) recipients has been shown safe and effective for the treatment of EBV-associated posttransplantation lymphoproliferative disorders (PTLDs). SOT recipients, however, require the continuous administration of immunosuppressive drugs to prevent graft rejection, and these ag...

Immunosuppressive Medications.

Immunosuppressive agents are commonly used in the nephrologist's practice in the treatment of autoimmune and immune-mediated diseases and transplantation, and they are investigational in the treatment of AKI and ESRD. Drug development has been rapid over the past decades as mechanisms of the immune response have been better defined both by serendipity (the discovery of agents with immunosuppres...

2-Amino-4-(nitroalkyl)-4H-chromene-3-carbonitriles as New Cytotoxic Agents

A series of 2-amino-4-(nitroalkyl)-4H-chromene-3-carbonitriles were synthesized by an efficient multicomponent reaction in aqueous media using DBU (1,8-diazabicyclo[5.4.0]undec-7-ene) as a catalyst at room temperature. Mild condition, environment friendly procedure and excellent yields are the main advantages of this procedure. The cytotoxic activity of target compounds were evaluated against t...

Synthesis, Biological Evaluation and Docking Analysis of Some Novel Quinazolin Derivatives as Antitumor Agents

Different acid chlorides (2a-d) reacted with anthranilic acid to produce 2-substituted-3, 1-benzoxazin-4-one (3a-d) which was used as starting material to synthesize some condensed and non-condensed heterocyclic compounds by reaction with nitrogen nucleophiles e.g., hydrazine hydrate, and formamide. Some of the newly synthesized analogues were chosen to evaluate their cytotoxic activity against...
